Bioo Scientific Launches Barcoded Adapters for Sample Multiplexing in Next Generation Sequencing

Bioo Scientific has introduced AIR™ Barcoded Adapters which allow sample multiplexing in small RNA (ie. miRNA ) Next-Generation Sequencing reactions, while decreasing costs and increasing the number of samples that can be run simultaneously. miRNAs are small, single-stranded RNAs that regulate gene expression by partial complementary base pairing to specific mRNAs. Recently it has been shown that these small RNAs regulate gene expression by targeting mRNAs for translational repression and degradation. miRNAs modulate many cellular pathways including growth and proliferation, apoptosis and developmental timing. Multiplexed Next-Generation Sequencing using Bioo Scientific’s AIR Barcoded Adapters will significantly speed up the discovery and analysis of these important molecules.

AIR Barcoded Adapters, designed to work with Illumina Next-Generation Sequencers, provide flexibility in high-throughput sequencing applications. They significantly increase scale and throughput while reducing costs by allowing the user to pool multiple library preparations into a single sequencing reaction. The AIR Barcoded Adapters enable the user to detect rare sample events amongst hundreds of samples, across several time points and even multiple genomes. Predesigned AIR Barcoded Adapters, are available in sets of 10, 20, or 30 and were designed based on uniform melting temperature (Tm) and sequences that have unique color space. Once attached to a sample, they may be pooled together and sequenced in a single flow cell run. Analysis post run allows the user to identify the sequence barcode back to specific samples. AIR Barcoded Adapters can also be used to multiplex Next-Generation Sequencing reactions for piwi RNA (piRNA), small nucleolar RNA (snoRNA) and other small, noncoding RNAs.

Targeted In Vivo siRNA Delivery

Bioo Scientific has introduced the patent pending T3™ Conjugation Kit for targeted in vivo delivery of siRNA or any other RNAi agent. The T3 Conjugation Kit can also be used for targeted in vitro delivery of siRNA in primary cells or cell lines, which have mixed cell populations. Bioo Scientific has built upon its expertise in RNAi delivery to create a technology that will offer customers the ability to effectively delivery siRNA or microRNA (miRNA) into any targeted cell or tissue type they are studying.

RNAi has revolutionized biology by allowing researchers to directly observe the effects of the loss of function of specific genes in mammalian systems. However, achieving efficient in vivo delivery of siRNA to the appropriate target cell has been a major hurdle in the advancement of using RNAi in gene function studies and as a therapeutic agent. This hurdle can now be easily overcome using the T3 Conjugation Kit.

The T3 technology functions by conjugating a T3 Carrier to an antibody that is specific to a receptor or other extracellular domain specific to the targeted cell or tissue type. The T3 Carrier-Antibody Conjugate can then be loaded with an RNAi agent such as siRNA or miRNA. The RNAi agent loaded T3 Conjugate may then be administered systemically or locally to an animal (or cultured cells) where it binds to and is internalized by cells recognized by the antibody. Once inside the cell, the RNAi agent is designed for release from T3 allowing silencing of its intended target. The T3 Conjugation Kit technology will propel the validation of in vivo work, leading to a better understanding of cellular pathways, the identification of novel drug targets, and the ability to more efficiently deliver RNAi agents as drugs.

According to Dr. Masoud Toloue, Senior Scientist at Bioo Scientific, “siRNA/miRNA delivery remains the largest hurdle for in vivo research application and all efforts to make RNAi a real therapeutic modality. With the launch of our T3 Conjugation Kit, both scientists and clinicians will have access to an antibody based siRNA/miRNA delivery vehicle that allows the user to choose any antibody and siRNA/miRNA combination for their research. For the first time, T3’s targeting technology enables users to concentrate RNAi mediator on their tissue/organ or tumor of focus while limiting systemic toxicity and significantly reducing current dose requirements. As the first commercial siRNA targeted delivery vehicle to market, T3 will advance and revolutionize the entire siRNA delivery arena.” This research was funded in part by grants from the National Science Foundation.

Sensitive Detection of Melamine Contamination

Today BIOO Scientific announced the expansion of its portfolio of food safety testing solutions with a new kit which improves the detection of melamine in food. Melamine contamination has recently been identified in infant formula and other milk-based products from China. Melamine is a dangerous, nitrogen rich organic compound that is illegally added to food and feed to increase the apparent protein content values of food products. Consumption of melamine causes highly insoluble melamine cyanurate crystals to form in the kidney, which damage renal cells and can result in renal failure or death. The recent establishment of a 1µg/g (1ppm) safety threshold for melamine in infant foods has led to an immediate need for sensitive melamine detection methods.

Bioo Scientific’s MaxSignal Melamine Enzymatic Assay Kit has been developed to meet the industry’s need for increased sensitivity. The MaxSignal Melamine Enzymatic Assay Kit is a rapid, inexpensive aqueous extraction method that is able to detect down to 0.2 ppm of melamine. Bioo Scientific’s Toxicity Assay Kits Increase the Efficiency of Drug Development

The DNA Assembler - Rapidly Construct Biochemical Pathways

Researchers at the University of Illinois at Urbana-Champaign have created the DNA Assembler, a novel way to rapidly construct biochemical pathways in yeast. The DNA Assembler incorporates homologous recombination to allow scientists to assemble entire biochemical pathways in a single step only requiring a simple DNA prep and a one-step yeast transformation. Advantages of the DNA assembler method include:

- Assembly of multiple-gene biochemical pathways in a single step.

- Most rapid and efficient approach to construct large recombinant DNA molecules.

It can be used to construct custom-designed DNA molecules not only on a plasmid but also on a chromosome with the potential to assemble a very large biochemical pathway or even a whole genome.The DNA Assembler represents a powerful tool in the construction of biochemical pathways for synthetic biology, metabolic engineering and functional genomics studies.
